Granzyme M (GZMM)
Granulozyme M is generally considered to be the effector molecule of NK cells expressing the killer target cells. Zhang shuren's group used 5-fluorouracil chemotherapy drugs to treat mouse kidney cancer tumor cells after the survival of slow-cycle cells, which demonstrated that these drug-resistant cells have the characteristics of tumor stem cells.
Furthermore, gene expression of drug-resistant tumor cells and wild-type tumor cells was analyzed, and it was surprising to find that granulozyme M was significantly expressed in these drug-resistant cells with the characteristics of tumor stem cells.
This is not consistent with what is usually learned. To prevent human error, they confirmed this fact by analyzing multiple mouse and human tumor cell lines, as well as a series of clinical tumor specimens. Furthermore, through enhancing and inhibiting the expression of granulozyme M gene, and in vitro and in vivo studies, it was found that the expression of granulozyme M in tumor cells was related to tumor stem cells, and was closely related to tumor invasion, metastasis and epithelial interstitial transformation (EMT).
This important discovery rewrote the biological function of granulozyme M, proposed new mechanisms of tumor invasion and metastasis, and provided new clues for the treatment of tumors.
