But it is limited by the toxic side effects associated with it. Therefore, researchers at the University of Washington in the United States said that their team built a synthetic protein. It mimics the effects of IL-2 and avoids the toxic side effects associated with IL-2, a synthetic protein that promises to be a powerful cancer immunotherapy.
Because there are two different types of IL-2 receptors in the cell, IL-2 tends to bind to receptors containing CD25. These receptors are expressed on regulatory T cells, and activation of these receptors leads to proliferation of immunosuppressive cells and potentially fatal pulmonary edema, hypotension, and capillary leakage. In order to maintain the anti-cancer efficacy of IL-2 while reducing its side effects, the university of Washington team used a computer model to build a synthetic protein called NEO-201 from structural elements of IL-2 and IL-15. The synthetic protein binds to receptors made of CD122 and CD132, but not CD25. Furthermore, NEO-201 has a higher affinity for CD122 and CD132 receptor complexes than natural IL-2 cytokines, and it is also very stable and will not lose its affinity at high temperature.
