The deduced 508-amino acid POTE14A protein has a calculated molecular mass of 57.2 kD. POTE14A has 3 cysteine-rich N-terminal repeats, 6 central ankyrin repeats, and a C-terminal helical region. The deduced 594-amino acid POTE15B protein has a calculated molecular mass of 67.1 kD. Compared with POTE14A, POTE14B has extra sequence within the ankyrin repeat region. The deduced 487-amino acid POTE14C protein has a calculated molecular mass of 54.6 kD. Compared with POTE14A, POTE14C has a C-terminal truncation. In vitro transcription and translation of the POTE14 variants resulted in proteins with molecular masses in agreement with their calculated masses.By partially sequencing clones obtained from prostate and testis cDNA libraries, they determined that the POTE14 gene is transcribed.
